Flash Method Development

Flash Method Development

Often thin layer chromatography (TLC) is used in conjunction with Flash Chromatography for method development and testing the purity of the fractions taken.

A TLC plate can be used to predict a flash chromatography run:

To develop a flash separation, several simple steps can be taken:

  1. Filter sample (if possible)
  2. For the first run use Solid Sample Introduction Module or Pre-Column (if possible). This will protect your main column until more is known about the sample
  3. Collect fractions every 0.5 column volumes
  4. Analyse every fraction — use TLC to determine if component is present and its purity
  5. Weaker solvents generally give better but slower separations (see Flash Solvent Table)
